Section 8.4 - Nucleic Acid Detection and Quantitation in Electrophoretic Gels and Capillaries

چکیده
Molecular Probes’ new generation of fluorescent nucleic acid gel stains — the SYBR Gold, SYBR Green I and SYBR Green II dyes — are by far the best high-sensitivity reagents for staining DNA (Figure 8.60) and RNA (Figure 8.61) in electrophoretic gels.1 These gel stains provide greater sensitivity with lower background fluorescence than the conventional gel stain, ethidium bromide. In addition, Ames testing performed by an independent laboratory has shown that SYBR Green I stain is significantly less mutagenic than ethidium bromide 2 (Figure 8.62).

Detection and quantitation of unlabeled nucleic acids in polyacrylamide gels.
A method for the detection and quantitation of unlabeled nucleic acids in polyacrylamide gels is presented. The technique is a variation on UV shadowing. The technique is based on the observation that some storage phosphor screens (Kodak in this case) are sensitive to UV light in the range where nucleic acids absorb most strongly. Nucleic acid present in a gel resting on the screen absorbs the ...
متن کاملElectrophoretic Heterogeneity of the Sedimenting Components of Arabis Mosaic Virus
Electrophoretic heterogeneity in preparations of arabis mosaic virus (AMV) was due to differences in net surface charge among the sedimenting components. Bottom component migrated more rapidly than top component in polyacrylamide gels and on cellulose acetate strips. When nucleic acid was removed from bottom component the nucleic acid-free protein shell showed sedimentation and electrophoretic ...
متن کاملCrystallization of biological macromolecules using agarose gel.
Gellified media prevent convection and crystal sedimentation, and provide an attractive growth environment for optimising biological crystals. Agarose gels are particularly easy to use and they are compatible with most of the common crystallization methods. They also offer new possibilities like counter-diffusion techniques. This paper gives a brief overview of their general properties and pres...
متن کاملSemi-automated, single-band peak-fitting analysis of hydroxyl radical nucleic acid footprint autoradiograms for the quantitative analysis of transitions.
Hydroxyl radical footprinting can probe the solvent accessibility of the ribose moiety of the individual nucleotides of DNA and RNA. Semi-automated analytical tools are presented for the quantitative analyses of nucleic acid footprint transitions in which processes such as folding or ligand binding are followed as a function of time or ligand concentration. Efficient quantitation of the intensi...
متن کاملEvaluation of Nucleic Acid Sequence Based Amplification (NASBA) and Reverse Transcription Polymerase Chain Reaction for Detection of Coxsackievirus B3 in Cell Culture and Animal Tissue Samples
Enteroviruses are the causative agents of a number of diseases in humans. Group B coxsackieviruses are believed to be the most common viral agents responsible for human heart disease. Genomic data of enteroviruses has allowed developing new molecular approaches such as Nucleic Acid Sequence Based Amplification (NASBA) for detection of such viruses. In this study, coxsackievirus B3 (CVB3) was de...
